- Portal was a 'trial'. Size and assets specifically kept minimal. Had no idea it would be this popular.
- Portal was a 'test bed', Portal 2 is a 'game'.
- GlaDOS returns as the antagonist.
- Chell returns, and we play as her.
- Portal 2 takes place a long time after the original. Hundreds of years, in fact. The complex has overgrown.
- Those orbs seen at the end of Portal are personality orbs. They've cornered off sections of the labs as their own, and are concerned about the decaying state of the complex.
- Portal gun remains unchanged.
- Physics will now be part of portal puzzle solving. EG: A vent that sucks air in can be combined with a portal to 'suck' from a different location.
- "Paint" mechanic, various liquid substances that cause some reaction. For example, in the demo shown an orange substance on the floor caused Chell to be propelled upwards. Using portals you can apply these 'paints' to other areas of the level for puzzle solving.
- Reflection cubes will allow Chell to reflect lazers for various puzzles.
- The co-op campaign will have you playing as two bipedal robots. They like to hold hands.
- Splitscreen for people playing on the console, and 'picture in picture' available for those playing remote co-op (eg: PC). This allows you to easily communicate with the other player and combine efforts for puzzle solving.

I played through Portal again, since it's been patched with extra content (spoilers if you don't want to know):
The closing shot is a few seconds longer, as Chell/you is carried away by some sort of robot. There are also radios now scattered throughout the facility, which, if you pick them up and carry them to the appropriate spots, receive a morse code signal (earn an achievement for getting them all). If you've been following the ARG going on, the community used these to uncover details of the sequel in the first place.

It's not a big change, but I guess Valve want to solidify the fact that Chell survives the ordeal and will be around for the sequel, although it's set hundreds of years later, and the personality spheres have blocked off parts of the facility, which is now overgrown. I noticed in my replay that there were four spheres - one appeared to do nothing but was a morality sphere; one was super-curious about everything; one recited a recipe for a cake; and the other was just really angry. If their personalities reflect the environments that they've set up around them, that could make things very interesting. Although I don't know how any of them survived being burned in the fire -- assuming it is even the same spheres.

How is Chell still alive if it's set 100s of years in the future? Is she cyborg'd?

Presumably she's been in stasis.

What I'm not sure about is how the Portal storyline fits into the HL2 storyline (if at all). Is the first Portal set hundreds of years before HL2, or the sequel set hundreds of years after? When did the Borealis launch?
(The unofficial prequel Portal: The Flash Version Map Pack shows it launching while you're escaping the complex, but that is unofficial.)
